Research Focus: Role of IH in regulating deep cerebellar nuclear output
Approach: Electrophysiology in vitro, pharmacology, immunocytochemistry, microscopy
Preparations: In vitro slice preparations, tissue sections
Below are short summaries of projects published from my research.
BSc Thesis research: I worked on projects in several labs at the University of Toronto that ranged from stem cell technology to electrophysiology. I completed two BSc honours theses dealing with genetic screening for early detection of bone cancer and the expression profiles of a chloride transporter in the zebrafish visual system.
Western blot analysis of tissue factor release Effect of volutrauma on the extrinsic coagulation cascade
Here we showed that the high tidal volume activates the extrinsic coagulation cascade in the newborn rat but not in the adult. Exposure to volutrauma for as short as 15 min significantly increased the clot time in newborns. We speculate that activation of the coagulation cascade is mediated by Tissue Factor (TF) release. This study brings valuable clinical insights into the use of mechanical ventilation in premature babies. PDF
MSc Thesis research: My current work deals with the properties and functional role of IH channels in DCN cells.
Biocytin-filled DCN neuron and MAP-2 labeled dendritic processes Model - Deep nuclear neurons
Deep nuclear neurons represent the final output of cerebellum after integrating afferent excitatory pontine inputs and inhibitory input from cerebellar cortical Purkinje cells. These cells spontaneously discharge at rest but can respond to Purkinje cell inhibition with a transient rebound burst discharge. My project involves the use of patch clamp recordings and stimulation of synaptic inputs to understand the regulation and functional role of burst output in response to physiological patterns of input.
